چکیده

Contraction of the guinea pig saphenous artery induced by stimulation of perivascular nerves consists of an adrenergic and a purinergic component. Neuropeptide Y (NPY) potentiated the neural responses significantly at low stimulation frequencies but not at high frequencies when the contraction was maximal. After blocking the adrenoceptors with phentolamine, significant potentiation of the purinergic component by NPY could be demonstrated. The potentiating effect ofNPY was abolished in the presence of nifedipine. After desensitizing the purinoceptors with a,38-methylene-ATP, there was no potentiation of the remaining adrenergic component by NPY. NPY also has no effect on contractions induced by exogenous norepinephrine but potentiates those induced by a,4-methylene-ATP. These results suggest that NPY potentiates neurally induced contraction in the guinea pig saphenous artery by a specific action on the purinergic response. (Circulation Research 1991;68:1401-1407)

Inhibition of the excitatory junction potential in the guinea-pig saphenous artery by ANAPP3.

Electrical stimulation of the perivascular nerves of the guinea-pig saphenous artery elicited excitatory junction potentials (e.j.ps) in the smooth muscle cells. These e.j.ps were inhibited by arylazido aminopropionyl adenosine triphosphate (ANAPP3), a specific antagonist for the P2-purinoceptor, suggesting that ATP may be the neurotransmitter.

Inhibition of late vein graft neointima formation in human and porcine models by adenovirus-mediated overexpression of tissue inhibitor of metalloproteinase-3.

BACKGROUND Autologous saphenous vein coronary artery bypass graft surgery is complicated by late graft failure due to neointima formation and subsequent atherosclerosis. Growth factors and metalloproteinases (MMPs) act in concert to promote neointima formation. Tissue inhibitor of metalloproteinase-3 (TIMP-3), an extracellular matrix-associated MMP inhibitor, uniquely promotes apoptosis of isol...

Interactive effects of homocysteine and copper on angiogenesis in porcine isolated saphenous vein.

BACKGROUND After coronary artery bypass grafting procedures with saphenous vein, there is a protracted elevation of plasma homocysteine and copper. These interact to elicit endothelial dysfunction through promotion of superoxide. It has been suggested that angiogenesis and the formation of a neovasa vasorum is important in mediating vein graft patency. A novel in vitro model of angiogenesis in ...
